MES is a solid white powder that is stored under standard temperature. Its molecular structure contains a morpholine ring and an ethanesulfonic acid group. The morpholine ring is a nitrogen-containing heterocyclic compound with high stability and inertness, showing no obvious toxicity or irritation to organisms. The ethanesulfonic acid group is a negatively charged functional group with good water solubility and buffering properties. MES can be used as a buffering agent and accelerator in various fields, including cosmetics, biomedicine, in vitro diagnostics, sealants, and forestry.

Applications of MES in the biomedicine (wound healing) industry.

Chronic wound healing is a complex, multi-stage biological process. Some traditional Chinese medicine powders or ointments that promote wound repair often have high water absorption, resulting in poor hydration at the wound site. This inhibits wound recovery and reduces medication absorption through the skin surface, leading to a longer treatment course and slower efficacy.

Hydrogels have shown distinct advantages in wound treatment by promoting the healing of damaged tissues in a hydrated environment. However, colloidal dressings, while providing a sterile and hydrated environment, do not significantly enhance wound healing, leading to prolonged use and higher recovery costs. To address this issue, patent CN117883623A introduces a wound dressing that promotes wound healing.

 

This wound dressing is composed of eight components: modified collagen, gellan gum, cellulose, crosslinking agent, clotting factor, composite traditional Chinese medicine extract concentrate, calomel, and white wax. The steps to prepare the modified collagen are as follows:

  1. Place purified collagen into sterile deionized water, stir until homogeneous, then raise the system temperature to 40-60°C. Adjust the pH to 8.0-8.5 using NaOH solution, then add succinic anhydride. After stirring and reacting for 1 hour, adjust the pH to 7 using hydrochloric acid. Centrifuge at 5000 rpm for 15 minutes, collect the supernatant, and dialyze using a 3.5 kDa molecular weight cutoff dialysis bag for 12 hours to obtain acylated collagen.
  2. Add MES and NaCl to sterile deionized water, and adjust the pH to 5.1 with NaOH solution to obtain an MES buffer solution.

  3. Dissolve the acylated collagen obtained in step 1, 1-ethyl-(3-dimethylaminopropyl)carbodiimide, N-hydroxysuccinimide, and dopamine in the MES buffer solution. Stir thoroughly at room temperature under an inert gas atmosphere for 24 hours, then freeze-dry to obtain modified collagen.

By treating collagen with succinic anhydride and dopamine, cationic residues such as lysine amino groups in the protein are replaced, increasing the protein’s surface negative charge density. This shifts the protein’s isoelectric point to a lower pH, allowing it to attract more water molecules through hydrogen bonding, thereby increasing hydrophilicity and solubility in water. This modification not only enhances skin absorption of the protein but also increases the viscosity of the modified protein, thereby improving the gel strength of the dressing.
