RNase A is an endoribonuclease that attacks at the 3’OHphosphate of a pyrimidine nucleotide. The sequence of pG-pG-pC-pA-pG will be cleaved to give pG-pG-pCp and A-pG. The highest activity is exhibited with single stranded RNA.

Application:
Ribonuclease A removes RNA from DNA plasmid preparations and protein samples. It is used for RNase protection assays, to remove unspecifically bound RNA, analyze RNA sequences, and hydrolyze RNA contained in protein samples, and purification of DNA.
Ribonuclease A from the bovine pancreas has been used in a study to assess the hybridase activity of human ribonuclease-1. It has also been used to investigate particle-based and monolithic columns for cation exchange protein displacement chromatography.
