Bovine serum albumin is a biochemical reagent widely used in various disciplines of biology and in the fields of medicine and pharmacy, for example, as a blocking agent in western blotting. Albumin in the blood mainly serves to maintain osmotic pressure, pH buffering, carrier function, and nutritional function; in serum-free culture of animal cells, the addition of albumin can provide physiological and mechanical protection as well as carrier function.

Function:

BSA is commonly used as a stabilizer in enzyme preservation solutions and reaction buffers to stabilize or modify enzyme activity, particularly when certain enzymes are unstable or exhibit low activity at low concentrations. When added to these solutions, BSA may function as a “protector” or “carrier,” significantly enhancing the activity of various enzymes. Enzymes that do not require BSA generally show no significant impact when BSA is added. For most substrates such as DNA, BSA can enhance the efficiency of enzyme digestion and enable repeated cutting. At 37°C, after 1 hour of enzyme digestion, BSA helps stabilize the enzyme, as many restriction endonucleases are only stable for 10-20 minutes or even less at this temperature in the absence of BSA. Additionally, BSA can bind to metal ions or other chemical substances in the buffer or substrate DNA that inhibit the activity of restriction endonucleases.

Bovine Serum Albumin (BSA) is widely used in biochemical experiments, such as Western blotting (WB), where its addition increases the protein concentration in the solution and acts as a protective agent for enzymes. It helps prevent enzyme degradation and nonspecific adsorption, reducing enzyme denaturation caused by unfavorable environmental factors like heat, surface tension, and chemical agents.
