DL-Dithiothreitol(DTT)
DL-Dithiothreitol is a commonly used reducing agent known as dithiothreitol (abbreviated as DTT), which has strong reducing properties. Its reducing ability is largely due to the conformational stability of its oxidized state six-membered ring (containing disulfide bonds).
The reducing power is influenced by pH and can only exert its reducing effect when the pH is greater than 7. This is because only the deprotonated thiol salt anion (-S–) has reactive activity, while the thiol (-SH) does not; the pKa of the thiol group is generally around 8.3.
